Objective To observe the effect of 1,25(OH)2D3 on the expressions of glomerular bone morphogenetic protein-7 (BMP-7) and transforming growth factor-β1 (TGF-β1) as well as renoprotective effects and its mechanism in uremic rats. Methods Uremic rats were induced by the excision of the right kidney and ligation of anterior branches of the left renal artery. Uremic rats were divided as follows: uremic control group (UC group), uremic +1,25(OH)2D3 group (UD group). A group of normal rats were served as normal control group(NC group). At 8 weeks, systolic blood pressure (SBP), serum calcium, phosphorus, 25(OH)D3, fibroblast growth factor 23(FGF23), 24 h urine protein (24 h UP), urinary podoctye (UPC) were measured. The expressions of BMP-7 and TGF-β1 mRNA in glomerular were determined by reverse transcription polymerase chain reaction, and proteins were detected by Western blot. Results SBP was significantly elevated in the UC group and UD group compared with that in the NC group, but there was indistinguishable from each other. Compared with the NC group, the increase in phosphorus, FGF23, 24 h UP, UPC and the decrease in calcium, 25(OH)D3 were observed in the UC group. Compared with the UC group, the levels of phosphorus, FGF23, 24 h UP, UPC of rats in the UD group were significantly lower, however, the calcium and 25(OH)D3 levels in UD group were significantly higher. BMP-7 mRNA and protein expressions were significantly decreased and TGF-β1 increased significantly in the UC group compared with the NC group. BMP-7 mRNA and protein expressions in the UD group were increased by 14.8%, 31.1% and TGF-β1 decreased by 35.7%, 43.9% compared with the UC group. Conclusions 1,25(OH)2D3 ameliorate not only the metabolism of calcium and phosphorus, but also increase the expression of BMP-7 and decrease the expression of TGF-β1, alleviate the excretion of proteinuria and urinary podocytes.
