Analysis of DNA fingerprints on different types of biological contamination of other pollens in Liangyou932

In this study, DNA fingerprints of 6 different types of biological contamination of other pollens in two-line hybrid rice Liangyou932 were constructed by 5 SSR primer pairs that had been screened. The products of PCR were detected by rapid silver-staining procedure in nondenaturing polyacrylamide gels. The results of analysis indicated that DNA fingerprints of 6 materials were different and the efficiency that these primer pairs discriminate these materials was also different. The DNA bands of4materialswere the sane as that of F_(1) hybrids amplified by RM 277. However the DNA bands of materials from 1 to 4 were different from that of F_(1) hybrids amplified by PM 287 and PM 302 respectively, which showed the best efficiency to discriminate these materials PM 302 was ultimately chosen as primer pairs to identify seeds purity of Liangyou932 because the specific bands were easily discriminated with that of F_(1) hybrids amplified by PM 302.
