A two-dimensional YACpooling strategy forlibrary screening viaSTSandAIu-PCR methods

We havedeveloped a simple two-dimensional YAC pooling strategy tofacilitate YAClibrary screening via STSandAlu-PCRapproaches. Themethodhasbeen implemented usingthehumantotal genomicYAC library ofOlsonandcoworkers, anditsvalidity tested byisolation ofmany chromosomes 19-and21-specific YACs.TheAlu-PCRapproach isnotable inthatitis hybridization-based, suchthat PCRprimer pairs donot needtoberepeatedly synthesized andtested foreach screening step.
