The invention discloses a preparation method of wood frog skin collagen micro-powder. The method comprises the following steps: with wood frog skin as a raw material, extracting collagen and subsequently dissolving the collagen in acetic acid by using a gentle solvent for electro-spinning without adding any high polymer to prepare the wood frog skin collagen micro-powder, wherein the particle size range of the micro-powder is 200nm-3000nm, and the micro-powder is small in particle volume, also uniform in particle size, safe, nontoxic and easy to be absorbed by the skin of a human body. Therefore, the micro-powder overcomes the shortcomings that in the prior art, the raw materials for extracting the collagen are complex, the safety is hard to be guaranteed, and the collagen is hard to be absorbed by human tissues because the collagen powder particle is relatively large in size. The wood frog skin collagen micro-powder prepared according to the method disclosed by the invention can be added into various biomedical nonwoven fabrics and make-up products as a bioactive raw material so as to play unique recovery and whitening functions.
