HLA-B27 subtyping by PCR-SSP method

Objective: To detect the HLA-B27 subtype and investigate the possible clinical implication of genotype. Methods: Subtyping for HLA-B27 of polymerase chain reaction with sequence specific primers (PCR-SSP) method was applied. A total of 38 HLA-B27 positive samples detected by serology were entered into the study. Containing 24 samples diagnosed ankylosing spondylitis (AS), 14 diagnosed non-AS. Results: Among 38 samples detected, 36 samples were found to be HLA-B27 positive, 2 samples were negative, error rate was 5.26%. Of these 36 samples, B*2704 was 77.78% (28/36 samples). There was no significant difference between the rate of B*2704 positive sample in AS samples and non-AS samples (χ2=0.321,P 0.05) .Conclusion: The dominating subtype for HLA-B27 of Chinese is B*2704, AS is not associated with HLA-B27 subtypes, PCR-SSP has proved to be a high-sensitivity, rapid and accurate technique.
