Influence of Yupao Decoction on Apoptosis and the Expressions of Bax and Bel-2 of Granular Cell in Mice with Experimental Autoimmune Premature Ovarian Failure

To discuss the influence of Yupao Decoction,a Chinese compound fumula on apoptosis and the expressions of Bax and Bcl-2 of granular cell in mice with experimental autoimmune premature ovarian failure(POF).Methods:The ovarian antigen was elicited with ovarian tissue of mature SD female rats.The model of POF was established in BALB/C female mice by multiple sites subcutaneous injection.The mice were treated with Yupao Decoction after 3 times of injection. Prednisone was taken as the positive control.Apoptosis condition of granular cells in mice of every group was observed with Terminal deoxynucleotidyl Transferase mediated dUTP nick -end labeling(TUNEL)method.The expressions of Bcl-2 and Bax in ovarian granular cells were analyzed with immunohistochemistry stain.Results:Compared with normal group,the content of Bax of ovary granular cells in the model group increased significandy.The content of Bax decreased in each Yupao Decoction dose group.Compared with normal group,the content of Bcl-2 of ovary granular cells in the model group decreased significantly.The content of Bcl-2 increased in each Yupao Decoction dose group. TUNEL method showed that the number of apoptosis of granulose cell increased significandy in ovary in model group.The number of apoptosis of granulose cell decreased significantly in ovary in each Yupao Decoction dose group.Conclusion: Apoptosis of granular cells is an important element in etiology of autoimmune premature ovarian failure.Yupao Decoction can cut down the content of Bax and rise up the content of Bcl-2 in ovary tissues of autoimmune premature ovarian failure mice.Yupao Decoction can relieve the apoptosis of granulose cells in the ovary.Yupao Decoction can restore function of ovary by inhibiting apoptosis of granulose cells.
