Solvent system for separation of isovalerylspiramycins in high speed counter-current chromatography

Bitespiramycin is a new antibiotic with multi-components fermented by transgenosis engineering bacteria.To separately prepare isovaleryspiramycins functioned as major component of bitespiramycin is significant in medicine analysis and quality control.High speed counter-current chromatography(HSCCC),as a new purification technology based on liquid-liquid distribution,provides a method for the separation of isovalerylspiramycins.The partition coefficients of isovalerylspiramycins in a series of solvent systems were investigated as an important element for selection.The effect of temperature and pH value on partition coefficient was studied at the volume ratio 10 ∶ 1 ∶ 5 ∶ 7 of N-hexane-ethyl acetate-ethanol-water to optimize the separation conditions.This solvent system is preliminarily used in HSCCC separation for isovalerylspiramycins,and the mass fraction of isovalerylspiramycin Ⅱ and Ⅲ obtained are 95.8% and 98.4%,respectively,which has good effect for the separation of isovalerylspiramycin Ⅱ and Ⅲ.
