Detection of expression level of 43 ku excretory-secretory antigen gene of Trichinella spiralis by RQ-PCR.

Specific primers and probes were designed according to the known sequence of Ts43 gene of Trichinella spiralis in GenBank and 18 S rRNA gene of T.spiralis was used as a reference gene for the normalization of the total RNA of T.spiralis in different developmental stages.The expression level of Ts43 gene was obtained by the fluorescence cycle threshold(Ct)line.A real time fluorescent quantitative PCR(RQ-PCR) method for detection of expression level of 43 ku excretory-secretory antigen(Ts43) gene was established and used to detect Ts43 gene expression level of T.spiralis in different development stages.The results showed that Ts43 was expressed in all developmental stages,however,the expression level in adult worms and newborn larvae were lower than those in other developmental stages.The expression level gradually increased in larvae from day 18 post-infection,and reached the peak on day 30 post-infection.Then the expression level gradually decreased,but in larvae on day 58 post-infection it was still higher than that in adult worms and newborn larvae.The results demonstrated that expression of Ts43 gene was significantly related with the formation of the nurse cell.
