CONFRONTO FRA TEST MOLECOLARI E ANALISI DELLA PRODUTTIVITA' DEL VIRUS DELL'HPV IN UNA POPOLAZIONE DI DONNE SALENTINE A RISCHIO

2009
AbstrA ct Introduction: The HPV detection is by complex molecular methods, but the viral productivity can be detected only by messenger RNA individua- tion. Material and methods: we examined 140 women at risk for HPV infection, searching virus by REAL-TIME PCR, sequencing and molecular hybridization, classifying virus by genotype or genotypes in case of co-infections. To detect the viral oncogenicity we utilized a new test based on NASBA technology, the NucliSENS EasyQ, that highlights the mRNA presence in E6 and E7 regions. results: on 52 samples HPV positive, 19 had one or more oncogenic genotypes of HPV (16, 18, 31, 33 e 45); comparison between genotypes detected by REAL TIME PCR and L1 gene hybridization (DNA test), by NASBA and mRNA hybridization and the genotypes detected by sequencing, promoted the DNA test as the most secure in genotyping detecting; on 15 samples of 19 the genotypes were correspondents, on 4 of 19 they were only partially and in 2 cases there was no correlation between the sequencing and the two methods. conclusion: the contemporary detection of DNA and viral mRNA shows the presence of active infection, by onco proteins E6 and E7 persistence, who increase the oncogenic risk in women.
