Multi-dimensional cell-rotation using multi-core fibers and wavefront shaping

We present a novel dual-beam optical trap for multi-dimensional cell-rotation. A paradigm shift for refractive index tomography is achieved through wavefront shaping using multicore fibers (MCF), resulting in adaptive phased arrays. The amplitude and phase of each core can be independently controlled, enabling the delivery of tailored light-field distributions to control the cell orientation in the region of interest. Using in situ calibration and on-the-fly manipulation enables precisely controlled cell-rotation around multiple axes in three-dimensions, leading to full spatial frequency coverage and overcoming of the “missing apple-core”. This upgrades the performance of the three-dimensional refractive index distribution measurement of the cell.
