The invention relates to an improved deep layer filter for a one-time biotechnology method. The method comprises a deep layer filtering device and a flocculating agent, wherein the deep layer filtering device consists of a porous deep filtering layer medium; and the porous deep filtering layer consists of cellulosic fibers, an inorganic layer and an organic wet strength agent. Compared with the prior art, the improved deep layer filter has the characteristics that by matching of the added flocculating agent and multiple special filter layers, a process for separating cell culture fluid is simplified, the production cost is reduced, the working efficiency is improved, cross pollution among molecule substances is avoided, and therefore, the improved deep layer filter can be used for biotechnology, medicine and a pharmaceutical production process.
