Effects of Probucol on Podocin and Nephrin Expression in Rats with Adriamycin-induced Nephrosis

Objective To study the effects of probucol on podocin and nephrin expression in rats with adriamycin-induced nephrosis.Methods A total of 36 SD rats were randomly divided into three groups:control group(CG group,n =12),nephrosis group(NS group,n =12)and probucol group(NSP group,n =12).The nephrosis models were established by caudal vein injection of adriamycin.Twenty-four h urine protein quantification was performed at the end of the forth week.Light microscope was employed to investigate the pathological changes of the glomerulus,Western blot was carried out to determine the protein expression of podocin and nephrin.Results Twenty-four h urine protein was significantly increased in NS group and NSP group compared with the CG rats(P 0.05),and less urinary protein was detected in NSP group than in NS group(P 0.05).Protein expression of nephrin and podocin was significantly decreased in NS and NSP rats than in CG rats(P 0.05),while protein expression of these two components was up-regulated in NSP rats compared with the NS rats(P 0.05).Conclusion Probucol may achieve the protective effect on adriamycin-induced nephrosis by relieving the podocyte injury.
