Downexpression of Krüppel-like factor 8 inhibits invasion of lung carcinoma cells A549

Objective To investigate the effect of Kruppel-like factor 8 (KLF8) on invasion ability of lung cancer cell line A549 and its mechanism.Methods KLF8 short hairpin RNA (shRNA) was transiently transfected into A549 cells,forming three subgroups:A549,Mock and sh-KLF8.The protein expression of KLF8 was detected by Western blotting.Invasion ability of A549 cells was evaluated by Transwell assay.Related protein of epithelial mesenchymal transition (EMT) was quantified by Western blotting.Results After transfection of A549 cells with KLF8 shRNA,the protein expression of KLF8 was down-regulated bu 90.3% (P < 0.01).Compared to mock group,the number of invasive cells was significantly reduced sh-KLF8 group [(87 ± 5) vs.(16 ± 7),P < 0.01).For related proteins of EMT,E-cadherin protein expression was up-regulated 4.9 fold change (P < 0.05),and N-cadherin (44.5%,P < 0.05) and Vimentin (62.1%,P < 0.01) protein expression was down-regulated.Conclusion Our results suggest that sh-KLF8 may inhibit the invasion ability of A549 cells probably via the EMT pathway.
