Human parathyroid hormone 1-34 peptide analogue capable of expressing in series and being released by synchronousacid hydrolysis, preparation method thereof and use

2008
The invention relates to the gene engineering field, and relates to a human parathyroid hormone 1-34 peptide analogue which can be tandem expressed and be synchronously acidically hydrolyzed and released, a method for preparing the same and an application of the human parathyroid hormone 1-34 peptide analogue for treating osteoporosis. The N end of the hPTH (1-34) analogue has a dipeptidase IV/CD26 recognition site which can ensure that the hPTH (1-34) analogue can be recognized and cut by the dipeptidase IV/CD26 after the hPTH (1-34) analogue enters a human body, thereby playing the effect of preventing and curing the osteoporosis of a natural hPTH. The method for preparing the hPTH (1-34) analogue comprises the following steps of: recombining plasmids; constructing a corresponding genetic engineering bacterium; preparing a concatemer of the human parathyroid hormone 1-34 peptide analogue; and acquiring the human parathyroid hormone 1-34 peptide analogue.
