Comparative analysis of Lactobacillus-specific molecular profile of intestinal microflora between normal and cirrhosis Bama miniature pigs by PCR-DGGE.

Objective To analyze and compare the richness,Shannon-Wiener index and evenness of Lactobacillus community in intestinal microflora of normal and cirrhosis Bama miniature pigs.Method Lactobacillus-specific Polymerase Chain Reaction-Denaturing Gradient Gel Electrophoresis(PCR-DGGE) analysis of intestinal microflora was performed.Total bacterial DNAs of the fecal samples were extracted and the total DNA samples subjected to Lactobacillus-specific 16S rDNA-based PCR-DGGE.The molecular DGGE profile was subjected to UPGMA and Principal Component analysis using Quantity One(Bio-rad) and origionab 7.0 software respectively.Result No remarkable differences were observed in the richness,Shannon-Wiener index and evenness of Lactobacillus in intestinal microflora between normal and cirrhosis Bama miniature pigs by statistical analysis(P0.05).Principal component analysis also indicated that cirrhosis did not alter the Lactobacillus community in intestinal microflora of Bama miniature pigs.Conclusion No remarkable differences was observed in the Lactobacillus-specific DGGE profile between cirrhosis and normal Bama miniature pigs.
