In vitro Multiplication of Landscape Garden Vine Allamanda violacea cv. Cherry Jubilee

An experiment was carried out in the Biotechnology-cum-Tissue Culture Centre, College of Agriculture, OUAT, Bhubaneswar in the year 2011 with the objectives to standardize the surface sterilization time, media supplements for shoot proliferation and root formation of a wonderful landscape woody vine Allamanda violacea cv. Cherry Jubilee which is quite difficult to propagate. For in vitro studies node explants (8-10mm size) taken from branches of A. violacea cv. Cherry Jubilee plants were surface sterilized with 0.1% HgCl2 for different time period such as 1, 3, 5, 7, 9, 12 and 15 mins under the laminar airflow and cultured on MS medium supplemented with 1.0 mg/l BAP along with 0.5 mg/l NAA for initial culture establishment. For shoot proliferation study, the explants were surface sterilized with the best treatment and cultured on MS medium alone or in combination with BAP (1.0 and 2.0 mg/l) along with NAA(0.5 and 1.0 mg/l) and IAA (0.1 and 0.2 mg/l). Then 8–10 cm shoots were cultured on MS medium alone or in combination with NAA (0.5, 1.0, 1.5 and 2.0 mg/l) and IAA (0.25, 0.5, 1.0 and 1.5mg/l) for root formation study for 30 days. The results revealed that 0.1% HgCl2 for 7 min was found to be most effective for sterilization of the single node explants of the Allamanda violacea cv. Cherry Jubilee, where the treatment recorded no contamination and death, higher percentage of aseptic culture (100%) and survival (100%), early callus as well as bud initiation and production of longer shoots (4.33 cm). MS medium fortified with BAP(2.0mg/l) and IAA(0.2mg/l) was found to be most promising combination for early and profuse callusing (8.33 days), shoot initiation (17.67 days) and production of longer shoot (10.27 cm) with more no of green colour leaves. A branch containing 10 nodes can produce 10 soots in in-vitro condition. Within a period of 30 days none of the explants developed root primordia hence further studies on this aspect is essential.
