Study on the qrimary biological characteristics of osteoblast of healthy people in vitro

Objective To investigate the primary biological characteristics of osteoblast of healthy people in order to provide the necessary preparation for the study on the biology and pathology of bone in vitro.Methods Diaphysis of extremities from four-month old normal fetus was digested and grew,establishing osteoblast system of healthy people.Its morphology was observed by optical microscope and SEM.Giemsa staining and histological observation was performed.ALP activity and bone gla protein synthesis in osteoblast were examined and compared with fibroblast.CollagenⅠ of osteoblast was investigated by immunological means.Results The results showed that the biological characteristics of osteoblast were steady.The content of ALP in osteoblast was much more than that in other cells,furthermore,which could synthesize a special Bone Gla Protein(BPG) that had been used as a marker for identifying osteoblast.In addition,normal osteoblast could only synthesize type Ⅰ collagen.Conclusion A osteoblast system of healthy people has been established,which can provide necessary theoretical basis for the study on the biology and pathology of bone in vitro.
