Protective Effect of Cerebrospinal Fluid Containing Qianxin Kaiqiao Decoction on PC12 Cell Injury Induced by Hydrogen peroxide(H_2O_2)

Objective:To observe the protective effect of cerebrospinal fluid containing Qianxin Kaiqiao decoction on PC12 cell injury induced by Hydrogen peroxide(H2O2)and provide clinical proof of the formula.Methods:Sprague Dawley rats underwent intragastric administration of Qianxin Kaiqiao decoction twice a day for three and a half days,the administration dose for rats was(7.9 g·kg-1).Thus the cerebrospinal fluid containing Qianxin Kaiqiao was prepared.The model of neurocytes damaged by H2O2 had been established by adding 200 μmol·L-1 H2O2 to culture medium of PC12 cells.The expressions of Bax mRNA,Bcl-2 mRNA and Caspase-3 mRNA were tested by immunohistochemistry in the PC12 cells.Results:The cell activity in cerebrospinal fluid containing Qianxin Kaiqiaofang decoction groups were increased,the expressions of Bax mRNA and Caspase-3 mRNA decreased and the expression of Bcl-2 mRNA increased in cerebrospinal fluid containing Qianxin Kaiqiao decoction groups,compared with comparison group,the difference was notable(P0.05,P0.01).Conclusion:Qianxin Kaiqiao decoction showed an apparent protective effect on neurocytes damaged by H2O2.
