Summary OB J E T I V E: G iven that anesthesia and surge ry are know n by their immu n o m o d u l at o ry effe c t s , we aimed to compare the immune response after 1 or 3 anesthetic ex p o s u res to s evo fl u rane in a mu rine model without surge ry. MAT E R I A L A N D M E T H O D S: Young adult male mice, n o n c o n t rolled for body confo rm at i o n , we re exposed 1 (gro u p S1) or 3 times (group S3) to 3% sevo fl u rane in ox y gen (1.2 m a x i mun alveolar concentration) for 40 minutes. Untre at e d animals we re used as controls. Th ree days after in vivo ex p o s u re to sevo fl u ra n e, the cellular composition of peri p h e ral blood and of the spleen we re studied. We also studied the in vivo response to ex ogenous (sheep red blood cells: SRBC) and endogenous antigens (heat shock pro t e i n s : H S P 65 and HSP 70) as well as biomarke rs of tox i c i t y. RE S U LT S: The number of leuko cytes in peri p h e ra l blood decreased in S3 animals, and the number of ly m p h o cytes decreased in both groups. B cells in spleen d e c reased only in the S1 gro u p , but an increased in vivo response to SRBC was seen in both S1 and S3 mice in c o m p a rison with the control animals. Re-ex p o s u re to s evo fl u rane led to a decrease in immu n og l o bulin G re sponse only to HSP 70. Plasma marke rs of liver and kidn ey function did not ch a n ge after anesthetic ex p o s u re. CO N C L U S I O N: C h a n ges in leuko cyte populations in peri p h e ral blood and in antibody - p roducing capacity occur after either a single ex p o s u re or rep e ated ex p o s u res to sevo fl u ran e. However no ch a n ges occur in biomarke rs of tox i c i t y.
