Effect of Rosiglitazone on Renal TGF-β_1 Expression of Doxorubicin-induced Nephropathy in Rats

Objective To investigate the protective effect of peroxisome proliferators-activated receptor gamma(PPARγ)agonist rosiglitazone(RSG)against doxorubicin-induced nephropathy in rats by regulating transforming growth factor beta1(TGF-β1)expression.Methods Thirty rats were randomly divided into the normal group,model group and RSG-treated group,ten in each.Total serum protein,serum albumin,serum creatinine,urea nitrogen,blood glucose,cholesterol,triglyceride and urine protein within 24-hour were detected.Results By the end of the experiment,hyperlipemia,hypoproteinemia and hyperproteinuria were observed in the model group.There were significant differences in TP,ALB,TC,TG and 24 hour urine protein between the model group and RSG-treated group,and nephrotic pathological lesion was ameliorated in RSG-treated group with increased expression of PPARγ and decreased expression of TGF-β1.Conclusion RSG has anti-fibrosis and protective effect on kidney by upregulating the expression of PPARγ and downgrading that of TGF-β1.
