Construction of DArT system in sugarcane

【Objective】A mature and stable sugarcane DArT(Diversity array technology,DArT) system was constructed to provide a new molecular marker method for genetic diversity analysis, genetic map construction and marker-assisted breeding in sugarcane. 【Method】Seven different sugarcane germplasms were used to optimize genomic complexity reduction method,develop DArT system and analyze their genetic diversity. 【Result】The optimum genome complexity reduction method was obtained based on the combination of Pst I/Taq I, and then DArT system was built which was used to analyze genetic evolution relations among the 7 materials. The genetic similarity coefficient of 7 materials ranged from 0.497 to 0.811 with an average of 0.569. At the genetic similarity coefficient value of 0.546, 7 materials were divided into 3 groups. In the first group,GT29, Badila and GXS85-30(Saccharum spontaneum) all belonged to Saccharum genus. Narenga porphyrocoma and Erianthus rockii were clustered into the second group. The third group included Miscanthus floridulus 84-8 and GXA87-36.【Conclusion】Sugarcane DArT system constructed in this study was effective, because the results of genetic similarity analysis using the system was consistent with the predicted genetic relationship of the tested materials.
