Experiment of Implanting ~(125)I Seed Radiation to Treat Nude Mouse with Prostatic Cancer(DU145)

To observe the effect of 125I particles implanting on the growth of Du-145(a common Prostate Cancer cell lines),fifty male nude mice were divided into five groups(each group 10),and DU145 cells were injected into the mice subcutaneously to construct the model.Five of the ten mice in each group were implanted in the tumors with 125I particles,and the other five were implanted with no-radiation particles as controls.Each group were give differential period of radical time and dose.Measuring the diameter of the tumors everyday,the volume of the implanted tumors is figured out and recorded it.After the mice were killed,the tumors were separated out and send for pathological and immunohistochemical detection.The implanted tumors in experimental group(125I)were obviously smaller than the controls.The results indicated that the 125I particles could inhibit the growth of DU145 implanted tumors.However,the difference among the experimental groups(144 h and 216 h)had no statistical significance(P0.05).The expression levels of some protein(Bax,Caspase-3,Caspase-9)in experimental groups' tumors were higher than the control groups.Furthermore,pathological results demonstrated that the differences of period of radical time and dose could affect the apoptosis of the implanted tumor cells.It is conclused that unremitting and small doses of 125I radiation can inhibit the growth of implanted tumors under nude mice's skin,and effects of the inhibition change with the certain factors just as period of radical time and radical dose。
