Study on biological characteristics of gp130 monoclonal antibody B-S12

Objective To study the biological characteristics of gp130 monoclonal antibody (McAb) B S12. Methods Cell count and 3H TdR incorporation were used to assess the cellular proliferation and growth of human multiple myeloma cell lines XG 1 and XG 2 stimulated by both B S12 and the Fab and F(ab′) 2 fragments. In addition, the influences of anther gp130 McAb B P8, IL 6 McAb B E8 and IL 6R McAb B R6 on the effects of B S12 on the proliferation of XG 1 and XG 2 cells were observed. Results B S12 stimulated proliferation of XG 1 and XG 2 cells, and supported their long term growth. But, the effect of B S12 was dependent on keeping its intact binding ability. B E8 and B R6 had no influence on the effect of B S12, while B P8 could synergize and augment its effect. Conclusion B S12 is a stimulating gp130 McAb, and play a similar role to that of IL 6.
