Influence of Basic Fibroblast Growth Factor by Local Application Method (Soak) on the Healing of Subcutaneous Vascular Net Free Skin Graft in Rats

Objective: To study the influence of basic fibroblast growth factor (bFGF) by local application methed (soak) on the healing of remove subcutaneous vascular net free skin graft in rats .Metheds: 10 SD rats,(ordinary experiment animals ,female and male ,body weight 275g~285g) randomly divided into two bundles. Two skin grafts of subcutaneous vascular net were cut off in the back respectively and then transplanted in situs.The upside skin grafts were used were as test group,free skin graft soak 10 minuts in bFGF, and downside skin grafts were used as control group, free skin graft soak 10 minuts in NaCl . After operation, the survival of skin grafts was generally observed by naked eye. the skin grafts were sampled to observe numbers of blood capillary and growth of granulation tissues with microscope.Results: The survival of skin grafts in the test and control groups were 95% and 72% respectively ( t =1.753, P 0.01) .Under the microscope, the surviving area of experiment bundle had a great deal granulation tissues, a large number newly born blood vessels, distinctly below muscular layer. There were significant differences in the numbers of blood capillary and growth of granulation tissues between the two groups ( P 0.05) . There were significant differences in growth of granulation tissues ( P 0.01).Conclusion: bFGF,as an important vascularization factor , has a distinc auxo-action to the surviving of removed free skin graft.but also the comprehensive effect on cell proliferation . Local application may accelerate new vascularization.There was an important clinical significance in rising rate of tissue graft.
