The effect of Nd:YAG laser on Porphyridium cruentum growth and extracellular polysaccharide production

2002
Porphyridium cruentum was irradiated with Nd:YAGl laser(wavelengh 1060m) by different time as treatment dose. After irradiated, the activity of cells, growth rate, biomass and the extracellular polysaccharides yield of Porphyridium cruentum were investigated to analyse Laser irradiated phenomenon. The experimental results showed that the lethally rate of higher irradiated does (20sec; 30sec and 40sec) preponderated over the low dose, as to be from 17% to 44%. The higher dose irradiated cells growth raptly in continue cultivation, growth rate K value increased 18%、23.4% and 15.3% respectively to that of CK. Physiological and biochemical characteristic of cells after Laser treated were determined. Among those, extracelluar polysaccharides yield improved most, by 50%-150% than that of CK. Pigment content was varied at different irradiated dose. But biomass of treatment cells incubatd 10 days have no prominent distinction from the control. All results revealed that Nd:YAG Laser can be a hopeful mutagen in Porphyridium cruentum breeding.
