Abstract : The goal of the MOLDICE program was to exploit ion channels for practical ion channel based sensing. Stochastic sensing provides a detection method that can deliver extremely low false alarm rates while still maintaining a high probability of detection. In order to develop a practical system that could be deployed in the field, the EBS led team addressed several key system parameters. First, to eliminate the previous need for vibration isolation for ion channel measurements, a stable bilayer platform was implemented. This platform uses a glass nanopore membrane device which contains a nanometer scale orifice that provides a solid support for a suspended lipid bilayer and subsequent protein insertion. To provide highly specific analyte detection, the alpha hemolysin protein pore sensor was genetically engineered. The (M113K)7 pore was incorporated into the final system and is able to detect ppb concentrations of DTPMPA in the presence of ppm concentrations of a TDP interferent. The EBS led team developed a prototype system that was portable for offsite demonstration of an ion channel based detection system. The program resulted in the successful demonstration of this prototype system at an offsite location in Arlington, VA on May 2, 2008.
