Binding studies of crystal violet on proteins

Abstract Binding studies of crystal violet on Human Serum Albumin (HSA) and Casein (CA) were carried out at 25°C. The binding parameters such as amount for maximum monolayer coverage ( x m ), cross sectional surface area, surface excess, binding equilibrium constant are determined from the Langmuir linear equation best fits. Under higher concentrations of HSA, bound CV induce HSA dimerisation. This effect is not seen for Casein. A substrate sandwich model is attributed for cascadic protein dimerisation. Ionic strength effect favours CV binding in both the systems.
