Estudio morfológico comparativo del trasplante dopaminérgico en diferentes ganglios basales

Fetal mesencephalic graft into dopaminergic denervated structures is one of the restorative strategies used in the treatment of Parkinson’s disease. In the present study, we compared the morphological features of dopaminergic grafts into the striatum, the subthalamic nucleus and substantia nigra pars reticulata. E-mail: lisis@neuro.ciren.cu dopamine lesions of the substantia nigra pars compacta were divided into three groups and received injections of 300 000 embryonic rat ventral mesencephalic cells in the striatum, substantia nigra pars reticulata, or subthalamic nucleus. Graft survival was assessed using tyrosine hydroxylase immunohistochemistry, 4 months after transplantation. Results. Intranigrally and intrasubthalamically grafted cells, unlike intrastriatally grafted ones, spread toward several surrounding areas. The dispersion of dopaminergic neurons was greater in the ones implanted into the subthalamic nucleus. In the three cases, TH-immunoreactive cells tended to cluster in the periphery of the grafts, with fewer cells observed in its center. A network of TH-positive fibers was observed within the graft itself and also extended into the host in all directions. Dopaminergic neurons grafted in the striatum showed more developed neuritic processes and larger cell bodies. Besides, we have found that the graft volume and TH-positive cell number were greater in the intrastriatal grafts than in the intrasubthalamic or intranigral grafts.
