Mechanism of purinergic action on glomerular permeability for albumin (692.3)

Purinergic receptors are expressed on cellular components of glomerular filtration barrier and their activation may affect glomerular permeability for albumin leading to albuminuria. We investigated mechanisms underlying purinergic action on glomerular filter permeability for albumin measuring convectional albumin permeability (Palb) in single isolated glomerulus based on video-microscopy method. Effect of nucleotides in vivo was investigated using osmotic pump implanted subcutaneously into Wistar rats. Cultured rat podocytes were used for analysis of albumin permeability, PKG dimerization and immunofluorescence of actin cytoskeleton. Various nucleotides (ATP, ADP, UTP, UDP) or non-metabolized ATP analogues (2-MeSATP, ATP-γ-S) increased Palb in time- and concentration-dependent manner in vitro. Effects were dependent on P2 receptors activation, nitric oxide synthase and cytoplasmic guanylyl cyclase.
