Autogenous Tissue Engineering Bone for Segmental Bone Defects in Rabbits

Objective:To investigate the effect of implantation of autogenous tissue engineering bone on the repairing of segmental bone defects in rabbits. Methods:The rabbit mesenchymal cells were cultured. The autogenous tissue engineering bone with CS-Gel/HA-rhBMP-2 was prepared. The bone mesenchymal stem cells were as seed cells,and were carried out with complex CS-Gel/HA-rhBMP-2. The establishment of animal models of bone defect was to amputate the middle of forelimb 7 mm length of the rabbit bone tissue. In experimental group (the right side of each rabbit),the implantation was CS-Gel/ HA-rhBMP-2-seed cell in the rabbit segmental bone defect models. In control group (the left side of each rabbit),the implantation was CS-Gel/HA. No internal and external fixation was done in two groups. The wound was sewn up layer by layer. The X-ray grade and histological score were evaluated after 4,8 and 12 weeks of treatment. Results:There were no significant differences in the X-ray grade and histological score between experiment group and control group after 4 weeks of treatment. However the X-ray grade and histological score were significantly better in experiment group than those in control group at 8 and 12 weeks after treatment (P 0.05). Conclusion:The autogenous tissue engineering bone can cure the rabbit segmental bone defects.
