Genomic Analysis of Effects of Gestational PFOS Exposure on Neonatal Death and the Nervous System Development in Rat

To investigate the potential mechanism of PFOS-induced neonatal mortality at the molecular level and to find out the interaction between PFOS-induced mortality and developmental neurotoxicity,the RatRef-12 Expression BeadChip was used to identify changes in gene expression at postnatal days(PNDs)1 and 7.The Gene Ontology for translation genes into relative functions and the pathway for graphical representations of gene interactions were both employed for further analysis of the expression data.There are 864 and 642 genes significantly affected by PFOS at PNDs1 and 7,respectively(p 0.05).Some of bioprocesses related to differentially expressed genes have been involved in the PFOS-induced mortality including:death,the central nervous system development,blood circulation,response to stimulus,bone development,response to hypoxia,heart process and regulation of pH.Results of this study indicate that the disruption of nervous system may lead to PFOS-induced neonatal mortality through the disruption of the brain blood and oxygen system and interference with the normal functional process of brain.
