İNFERTİL BİREYLERDE SEMEN HACİM VE SPERM UZUNLUK ÖLÇÜMLERİNİN EZRİN VE N-KADERİN PROTEİNLERİ İLE İLİSKİLENDİRİLMESİ Association of Semen Volume and Sperm Length Measurements with Ezrin and N-Cadherin Proteins in Infertile Men

OZET Amac: Infertil bireylere ait sperm hacim/uzunluk oranlari ve spermiyogenezin fizyolojik surecinde aktin baglayici protein olan ve bir capraz baglayici olarak kortikal hucre iskeleti ile plazma membran proteinleri arasinda bulunan ezrin, ile bir transmembran glikoproteinlerin olan N-kaderin kolakizasyonlarinin arastirilmasi gerceklestirilmistir. Gerec ve Yontemler: N= 18 infertil bireyde hacim/uzunluk oranlari ve sperm sayimi gerceklestirilen semen onekleri sperm yayma preparatlari Kruger Kriterlerine gore mikroskop altinda incelendi. Her grup icin sperm uzunluk olcumleri gerceklestirildi, immunofloresan boyama ile ezrin ve N-kaderin lokalizasyon ve ekspresyonlaridegerlendirildi. Bulgular: Sperm uzunluklarinin hacim yogunluklarina oranla azaldigi ve sperm uzunlugunun sperm sayisi ile arasinda anlamli korelasyon oldugu tespit edildi. Sperm kuyruk uzunlugu anomalisinin infertilite uzerinde etkin bir durum oldugu ancak ezrin ve N-kaderin lokalizasyon ve ekspresyonlarinda control grubuna gore anlamli fark olmadigi tespit edildi. Sonuc: Sperm sayisi/hacim ve sperm uzunluk oranlarinin infertil bireylerde spermiyogenezin fizyolojik surecine bagli olarak gelisebilecegi ilk kez bu proje ile gosterilmis olup, sperm morfolojik ozelliklerinin semen hacmi ile iliskili olabilecegi ve infertil bireylerde belirli fonksiyonel proteinlerin tespitinin gerceklesmesine yonelik calismalarin gerekliligi vurgulanmistir. Anahtar Kelimeler: Erkek infertilitesi; Semen analizi; Sperm uzunlugu; Ezrin, N-kaderin ABSTRACT Aim: The aim of this study was to investigate the sperm volume/lenght ratios of infertility individuals and N-cadherin co-localizations, which are the actin binding protein in the process of spermiogenesis, and ezrin, which are between the cortical cell skeleton and plasma membrane proteins, as a cross-linker. Material- Method: Semen samples taken from infertilite individuals (N=18) examined by microscopy after staining and fixation. The volume/length ratio was performed. Sperm morphology was examined as sperm smear preparations according to Kruger Criteria. Sperm length measurements were performed for localization and expression parameters of ezrin and N-cadherin were examined by immunofluorescence staining. Results: The sperm length of the patients with similar morphological criteria decreased with respect to volume and there was a significant correlation between sperm length and sperm count. It was found that sperm tail length anomaly was effective on infertility, but there was no significant difference in localization and expression of ezrin and N-cadherin compared to control group. Conclusion: It has been shown for the first time that sperm count/volume and sperm length ratios may develop depending on the process of spermiogenesis in infertile individuals, it is emphasized that sperm morphological features may be related to semen volumes and necessity of studies fort he detection of certain functional proteins in infertile individuals is emphasized.
