Optimization on SSR analysis system of Pyrus L.

The factors that the concentration of DNA template,the concentration of dNTP,the concentration of Mg2+ and Taq DNA polymerase and the effects of annealing temperature on SSR analysis on pear were studied,and the optimized SSR system for pear was established.The optimized SSR system for pear was as following: in 20μL volumes SSR reaction system,30ng of DNA template,1.2mmol/L of Mg2+,0.15mmol/L of dNTP,1.0U of Taq DNA polymerase,0.8μmol/L of primer and the annealing temperature of primers of 49-55℃.Nineteen SSR primers that were suitable for pear were screened.Polymorphism between different cultivars was abundant and stable by 8% polyacrylamidegel,which showed this system was suitable.
