The invention discloses an automatic dot matrix positioning sheet punching apparatus. The apparatus comprises a base, a sheet punching mechanism, a moving mechanism, an air compressor blowing device and a control device for controlling the mechanisms, wherein the moving mechanism is fixed on the base and provided with a moving platform for placing an elisa plate; the sheet punching mechanism is positioned above the moving mechanism and provided with a guide tube for guiding a blood round sheet to fall into the holes of the elisa plate below; and the control device controls the position movement of the moving mechanism to be matched with the sheet punching action of the sheet punching mechanism and the blowing action of the air compressor blowing device.
