Effects of resveratrol on the release of ATP in cultured astrocytes with stretch injury.

Objective To study the effects of resveratrol on the release of ATP in cultured astrocytes with stretch injury and the protective effect of resveratrol on the central nervous system. Methods Astrocytes were treated with various concentrations of resveratrol for 12 h. The levels of ATP and lactate dehydrogenase (LDH) leakage were examined after stretch injury. Results ATP assay showed that stretch injury caused an increase in ATP level in the culture media. At the concentration of 1 μmol, resveratrol obviously increased the level of ATP, whereas 100 μmol of resveratrol inhibited the release of ATP. LDH leakage assay showed that stretch injury also caused the increase in LDH leakage in astrocytes. 1 μmol of resveratrol was found increase LDH leakage, whereas 100 μmol of resveratrol inbibited LDH leakage. Conclusion Resveratrol could inhibit the release of ATP and LDH leakage and protect astrocytes after stretch injury.
