Influence of Tangnaikang Formula on glycolipin metabolism and kidney morphology in type 2 DM KKAy mice

Objective: To investigate the effects of Tangnaikang on renal pathological changes in the early period of type 2 diabetic KKAy mice model.Methods: 12 weeks' male KKAy assigned into five groups randomly: diabetes(DM) group(n=10),diabetes with high Tangnaikang therapy group(n=10),diabetes with middle Tangnaikang therapy group(n=10),diabetes with low Tangnaikang therapy group(n=10),diabetes with valsartan therapy group(n=10) and C57BL/6J mice.Body weight and FBG was detected once of 2 weeks,24h urine protein quantitative was detected once of 4 weeks.After 8 weeks,All the mouse were sacrificed and the blood serum was obtained,TG,TC,Bun and Scr were detected;Early renal pathological variation was presented by histochemical staining.Kidney FN mRNA expreesion was detected by realtime fluorescence quantitative PCR method.Results: In DM group mice,renal glomerulus area was magnified,GBM was thickening and obvious protein casts were observed compared with those of normal group.Body weight,kidney weight,4h urine protein quantitative,FBG,INS,TG,TC and kidney FN mRNA expreesion was increased.After therapy for 8 weeks,Tangnaikang treatment can alleviate these pathological changes.Conclusion: The results indicate that Tangnaikang can ameliorate the renal pathological variation in the early period,by its effects of decreasing blood glucose and serum lipids.
