Experimantal and clinical research of Interferon on bladder cancer cell line T24 and patients

OBJECTIVE:To investigate the effects of different concentrations of interferon-α(IFN-α) on human bladder cancer cell line T24 and on 25 patiants with bladder cancer. METHODS: The cytotoxicity effect of IFN-α was assayed by MTT. Apoptosis was measured by flow cytometry,TUNEL and electron-microscopy. Twenty-five cases were treated with local injection of IFN-α before operation. RESULTS: Low-dose IFN-α did not have any significant effects on growth of T24 cells.High-dose IFN-α (10 000 U/mL) induced apoptosis and had significant cytotoxicity effects with dependence of time and dosage on T24 cells (F=84.544, P=0.007). Tumor size became small obviously after the injection (F=83.257, P=0.009). CONCLUSIONS: High-dose IFN-α (10 000 U/mL) has significant cytotoxicity effects on T24 cells and induces apoptosis of bladder cancer cells. Local injection of IFN-α may become one of effective therapy for bladder cancer.
