RP-HPLC Determination of Indapamide in Human Whole Blood

Objective:An RP-HPLC method was developed for the determination of indapamide in human blood. Method:The chromatographic separation was carried out on Dikma Diamonsil C_(18) column (200 mm×4.6 mm, 5μm)with the mobile phase consisting of 0.1% triethylamine buffer(adjusted pH to 3.75 with phosphoric acid)- acetonitrile(65:35),flow rate at 1.0 mL·min~(-1).Detection wavelength was set at 240 nm and column temperature was 40℃C.Carbamazepine was used as the internal standard and blood samples were extracted by ether.Result:The linear range was 5-1000 ng·mL~(-1),r=0.9999.The method recovery was within 94%-103%.Within-day and between-day RSD were less than 7%.Conclusion:The method is sensitive,specific and reproducible for the de- termination of indapamide concentration in human blood.It is very suitable for pharmacokinetics and bioavailability study of indapamide.
