Identification of fatty acid conjugates of plaunotol, a new anti-ulcer agent, formed in rat liver homogenate system.

Plaunotol, a new anti-ulcer isoprenoid (C20) which possesses two hydroxyl groups at the 1- and 18-positions, was readily conjugated with fatty acids by esterification in a rat liver homogenate system without added cofactors. The hydroxyl group at the 1-position was exclusievly esterified in vitro. The fatty acid conjugates of plaunotol served as substrates for cholesterol esterase. After purification by thin layer chromatography and high performance liquid chromatrography, esters with palmitic, oleic and stearic acids were identified by comparing the gas chromatography-mass spectrometry with those of synthesized authentic standards.
