Molecular Identification of New Wheat Cultivar (Chuannong) 16

In order to utilize gene resource efficiently, the genetic variations among new wheat cultivar (Chuannong) 16,its parents (i.e. Chuanyu 12 and 87-429) and other cultivars (i.e. Chuanmai 28 and Mianyang 26) were analyzed and identified by RAPD, STS and SSR markers. The results indicated that the genetic (differences) among (Chuannong) 16 and its parents were detected and all materials could be distinguished by the three kinds molecular (markers). 68 polymorphic bands were observed in 22 of 67 random primers (32.8%) in RAPD analysis, Genetic (differences) were revealed in 6 of 12 primer-sets (50%) and 21 of 120 (primer-enzyme) combinations (17.5%) in STS analysis, and in 11 of 34 SSR primer-sets (32.4%) in SSR analysis. In this study, the results suggested that SSR (markers) combining with (polyacrylamide-)(gel-electrophoresis) were superior to RAPD and STS markers in wheat cultivar identifying and fingerprinting.
