Influence of the time of middle cerebral artery occlusion on induction of cerebral ischemia-reperfusion injury in mice

Objective: To investigate the influence of the time of middle cerebral artery occlusion(MCAO) on cerebral ischemia-reperfusion injury in mice.Methods: In male C57BL/6J mice,MCAO was performed for 0.5 h,1 h and 2 h.Neurological deficits,survival rate,the infarct volume and brain water content were evaluated 24 h after reperfusion.Results: Ischemic mice exhibited brain damage after MCAO.One-hour ischemia and 24-hour reperfusion resulted in significant neurological deficits,and presented much aggravated infarct volume and brain water content compared with sham group,with the minimal variation coefficient.Conclusion: One-hour occlusion and 24-hour reperfusion can induce typical cerebral infarction in mice,with suitable size and easy reproduction.Therefore,it may be beneficial for physiological and pathological study,and pharmacodynamic evaluation for brain ischemia.
