Detecção de fraude em amostras comerciais de queijo bubalino por adição de leite bovino por meio da técnica de Reação em Cadeia da Polimerase (PCR) multiplex

This study aimed at verifying the efficiency of the technique of multiplex Polymerase Chain Reaction (PCR) for detecting the fraud by adding bovine milk into cheese samples made from buffalo milk, and to identify the fraudulent cheeses samples marketed in the municipalities of Para and Amapa. The buffalo cheeses samples containing 10 %, 25 %, 50 %, 75 % and 90 % of bovine milk were experimentally produced; and also the cheeses made from the cow and the buffalo milk were separately prepared as controls. The analysis of these samples was performed by multiplex Polymerase Chain Reaction. Subsequently, 44 samples of buffalo cheeses were collected from the study target region, and they were analyzed by PCR for detecting the fraudulent specimens. The primers used in this study showed actual performance for detecting all of the increasing percentages of cow milk experimentally added into buffalo cheeses; also it indicated that 13.6 %of these marketed samples contained bovine milk. It was concluded that the multiplex PCR is a suitable technique for detecting the fraud from the concentration of 10 % bovine milk added into buffalo cheese.
