Bioinformatics of Differentially Expressed Genes Related to Dimorphic Transition of Sporothrix schenckii

2011
Objective To investigate the molecular mechanism of dimorphic transition between mycelium(M) and yeast(Y) phases of Sporothrix schenckii, and screen the differentially expressed genes related to the transition. Methods The cDNA subtractive library between M and Y phases of S.schenckii was constructed by suppression subtractive hybridization(SSH) technique,in which the differentially expressed genes were analyzed for bioinformatics. Results A total of 751 and 875 expressed sequence tags(ESTs) were obtained from M + Y and Y + M libraries, with which 101 and 249 unigenes were obtained by splicing, respectively. The dimorphic transition between M and Y phases was accompanied by high expression of differential genes which were classified as structural gene as well as the genes encoding metabolic enzymes, cell surface molecules and the molecules with indistinct function. Conclusion The cDNA subtractive library related to dimorphic transition of S.schenckii was successfully constructed, from which partial differentially expressed genes were screened, which laid a foundation of further study on the genes related to the pathogenicity of S.schenckii.
