Whole Cell Dry-coating in ELISA for Detecting Serum Antibody against Fowl Cholera

Pasteurella multocida was integrally dry-coated on the basis of glutaraldehyde coupling of the whole cell antigen to microliter plates.Glutaraldehyde of 2.5% concentration was used for pretreatment of ELISA plates.Using the method of chessboard titration,a hundred million of whole cells per milliliter in PBS buffer were selected for the proper dry-coating concentration.Specificity,repeatability and stability examination's result as well as the preliminary indirect ELISA used in the sample test indicated that the whole cell dry-coating could be rather helpful to development of ELISA detecting fowl cholera antibody.
