Accelerated repair of acetic acid induced gastric ulcers using antiviral and antibacterial vaccines stimulating angiogenesis and fibrogenesis in rats

Objective: To stimulate the angiogenesis and fibrinogenesis in gastric ulcers (GU) produced by glacial acetic acid (GAA), utilizing antiviral and antibacterial vaccines, and compare the ulcer repair with the one produced by basic fibroblast growth factor (bFGF). Method: Groups of Wistar rats (n=42), weighing 200g, 24h fasting, provided with water ad libitum, and anesthetized were submitted to laparatomy. 0.025ml of GAA was injected in the mucosal gastric subserosa, at the junction of the antrum and the gastric body. At 24 hr every group (n=7) were submitted to a second laparatomy and reinjected in the same region. Group 1:0.5 ml saline was administered and the laparatomy was closed. G 2:100 ~g/Kg bFGF. G 3: 0.5ml influenza vaccine (Pasteur M~rieux). G 4:0.5 ml triple vaccine (D.T.Coq/D.T.P. Pasteur M~rieux). G 5: 0.5ml measles vaccine (Morbilvax-Biocine S.P.A.). G6." 0.5 ml polio vaccine (SB Biologicals Rixensart). The groups were evaluated on day 7. Results: Maroscopy. The ulcer area was measured (mm2). G 1:6.5 + 1.5; G 2: 2.0+_0.5; G 3:0.5+_0.1 (p<0.05); G 4:0 .0 (p<0.01); G 5:0.5+_0.1 (p <0.05); G 6:0 .0 (p < 0.01). Histological examination: G 1: extensive ulceration with mucosa, submucosa and muscular layer necrosis. Mild reactive proliferation of fibroblasts and endothelial cells. G 2: Small mucosa necrosis and important fibroblast and miofibroblast proliferation with important angiogenasis. G 3: Minimum central focal mucosal erosion with mild reaction of fibroblasts and endothelial cells. G 6; No ulceration was observed. Intensive reactive proliferation of fibroblasts and endothelial cells. Conclusions: 1) The important proliferation of fibroblasts and endothelial cells and the great angiogenesis stimulated by bFGF, accelerated the GAA gastric ulcer repair. 2) The ulcer repair stimulated by triple and polio vaccines was more rapid and associated with an important proliferation of fibroblasts and miofibroblasts, and a great angiogenesis. 3) Both measles and influenza vaccine were able to accelerate significantly the GAA gastric ulcer repair in comparison with the controls.
