Oil palm tender leaf minimal damage material fetching and callus tissue induction method

2015
The invention belongs to the technical field of plant tissue culture and relates to an oil palm tender leaf minimal damage material fetching and callus tissue induction method. The method comprises the following steps: holing on a longitudinal upward part of a growth point away from a mature oil palm, taking out tender leaves, disinfecting, cutting into small pieces and inoculating on a callus tissue inducing culture medium to culture, wherein the culture conditions are as follows: the dark culture is adopted, and the culture temperature is 26-30 DEG C; after 45-60 days of induced culture, starting to generate callus tissues. The oil palm tender leaf minimal damage material fetching and callus tissue induction method has the advantages that the process is simple, the operation is convenient, the tender leaves are collected through holing on the growth point, the damage to an explant seed tree is very less, the recovery after harvesting is quick, the treated leaves are inoculated onto the callus tissue culture medium, the callus tissues with higher induction rate and good quality can be obtained, a good foundation is laid for an efficient oil palm somatic embryo regeneration plant technology system, and the realistic significance in the protection, the development and the utilization of oil palm excellent germplasm resources is realized.
