Process Optimization for an NS0-Derived Hybridoma Cell Line in a Chemically Defined, Protein-Free Medium

A process optimization study was performed in stirred-tank bioreactors to determine parameter set points that maximize cell growth, viability, and monoclonal antibody (MAb) production. A statistical design of experiment (DoE) software package was used to determine parameter settings for dissolved oxygen (DO), pH, and temperature. An immunoglobulin G (IgG) producing NS0-derived hybridoma cell line was adapted to growth in a chemically defined, protein-free and animal-component free medium. Growth and productivity were characterized at the 2L bioreactor scale. Results were analyzed based on variable responses including growth and productivity on a per-liter basis. Parameter ranges that yield optimal productivity were determined.
