Determination of amikacin by fluorescence spectrophotometry

Amikacin has weak fluorescence at an excitation wavelength of 295nm,and an emission wavelength of 367nm.In the presence of sodium nitrite,amino-group was converted to nitryl.and then hydrolyzed in acidic conditions to produce reducing sugar.The reaction of reducing sugar with ethylenediamine in phosphate buffer solution will produce a strong fluorescent substance.A new method of measuring the content of amikacin was established according to this.A linear relationship between the concentration of amikacin in the range of 2.0×10-5-5.0×10-6 mol/L(r=09952) and fluorescence intensity was obtained with the detection limit of 4.2×10-7mol/L.This method was applied to the measurement of amikacin sulfate.
