RAPD analysis of genetic diversity in four different populations of rotifer Brachionus plicatilis

RAPD technique was applied to evaluate genetic diversity of four different rotifer(Brachionus plicatilis) populations which were collected from wild eggs in Dalian,Panjin,Dongying,Tianjin and cultured under the lab conditions.DNA of rotifers were extracted by frozen method and amplified DNA under the RAPD optimized conditions.In this study,amplification with 30 random primers produced 311 clear and reproducible stripes from 26 random primers.The length of amplified fragment was ranged from 190 to 2000 bp.According to a index of stripes sharing,Nei distance and clustering analysis among four populations the heredity distance between Dalian and Tianjin populations was the maximum.On the contrary,the heredity distance between Tianjin and Panjin populations was the minimum.Using UPGMA clustering analysis the Panjin and Tianjin populations were clustered into one group,followed by Dongying and Dalian populations.
