Immune reactivity between the stage-specific antigenic components from Schistosoma japonicum and the immune sera from rabbits vaccinatedwith irradiated cercariae

2005
To screen the antigenic components with protective immunity by comparing the differences of the stage-specific antigens from Schistsoma japonicum and their immune reactivities with the immune sera from rabbits immunized with irradiated cercariae, the mature and immature egg antigens, cercariae antigen, female and male adult worm antigens were collected and purified respectively. The antibody levels in sera of rabbits infected with either the normal or with the irradiated cercariae were detected by ELISA, and the immune reactivities of these sera with different stage-specific antigenic components were analyzed by means of SDS-PAGE and Western blotting assay. The experimental results revealed that significant differences in the antigenic proteins existed (among) the mature and immature egg antigens, cercariae antigen, female and male adult worm antigens as demonstrated by SDS-PAGE.The results revealed by Western blotting indicated that 23 antigenic components from the above mentioned 5 stage-specific antigens could be recognized with high immunological reactivity by the immune sera of rabbits immunized with irradiated cercariae. The molecular weights of these 23 antigenic components were 24.5?kDa, 28?kDa, 36?kDa, 43?kDa, 44?kDa, 48?kDa, 54?kDa, 58.5?kDa, 60?kDa, 62?kDa, 66?kDa, 68?kDa, 70?kDa,75?kDa, 79?kDa, 85?kDa, 87?kDA, 91?kDa, 95?kDa, 97?kDa, 105?kDa, 108?kDa, 110?kDa respectively.
